Spiceheads,
I’m back, at a new job, in a new environment with new challenges!
I’ve inherited an Office 365 environment that lives in the cloud. Meaning that it’s not sync’d or related back to the DC on premise. This is causing issues where I can’t manage the Cloud AD through the on-prem AD. Meaning any change has to be manually copied over to the other environment.
Now, the issue - or questions rather that I have is; can I simply start DirSync on the DC and the accounts will magically match up? Or will that drop the 400GB+ of email that’s stored in the Exchange Online arm of Office 365?
Any further information needed - just ask,

Do you still have a local Exchange Server (maybe hybrid mode) ?

If your users’ UPNs match the name in Office 365, it will match them up and no data loss will occur.
For instance, users in Active Directory have UPN of user@company.com and the naming convention is the same in Office 365. If so, once DirSync is configured, it will match the AD account to the Office 365 account.
Does on-premise has only AD or hybrid Exchange ? If you have only AD, you can sync freshly. Does AD connect installed already on the DC and what is the status of that sync ?
This may be a minor terminology point, but Dirsync is really old, you should be on AADConnect these days. If that’s what you meant, cool.
I’ve never tried populating an on-prem DC with existing cloud accounts, it’s always been in the other direction for spinning up a new O365 tenant from an on-prem AD.
